Density gradient centrifugation of diluted whole blood on a polysucrose-sodium metrizoate medium is the method of choice for isolation of lymphocytes. The LeucoVacTM system allows the blood sample to be drawn directly into the centrifuge tube with no special precautions required to prevent disruption of the polysucrose-sodium metrizoate layer.
